Is pH condition change after synthesis of chitosan coated silver nanoparticle?
During synthesis of chitosan coated silver nanoparticle the Zeta value shows negative charge (-39), after store in 4 degree Celsius the the Zeta value showing positive charge (+50). Is it because of pH ? If it because of pH how could i store my sample?

I am getting smear on gel after using linear DNA (amplicon) as a template while this is not the case with circular DNA (plasmid) as a template with same set of primers. I have used various amounts of template, gradient PCR, lowered the amount of polymerase; but, no success. I am using Phusion™ High-Fidelity DNA Polymerase Kit from Thermo. I will highly appreciate your input.
